Thanks I think he is ok and I have started shutting out the light. I am going to try to send a picture of the spot on his comb but don't know how well you can see it.

ron. first turn off the light. this is what egg production houses do to make the hens lay 24/7 thus shortening thier lives, as the never get to sleep. let them sleep ... you will have happier girls. get some diatomaceous earth and mix some into the food occasionally, spread some in thier dust baths. leave the chemicals alone.
i wouldnt worry about the black spots on the comb unless you have other symptoms. its likely mite bites or been peck or as someone said frost bite. likely harmless.
